Abstract

Acetone-Butanol-Ethanol (ABE) fermentation using Clostridium acetobutylicum is one of the main pathways for biobutanol production from lignocellulosic biomass and industrial sugar sources such as molasses. However, the success of ABE fermentation is highly sensitive to several technical factors, and failure to meet these conditions may lead to complete fermentation failure. This study aimed to evaluate and identify the critical factors causing failure in ABE fermentation using molasses media at concentrations of 10, 20, 30, 40, and 50 °Brix. Fermentation was conducted at 37°C, pH 6.5, with 10% (v/v) inoculum for 14 days under anaerobic conditions using tightly sealed Erlenmeyer flasks equipped with rubber stoppers and outlet tubing connected to a water-lock system. The results showed no production of biobutanol, acetone, or ethanol at all concentration variations, as confirmed by GC-FID analysis. The absence of gas bubble activity (CO₂ and H₂) from the first day and the stable pH values throughout fermentation indicated bacterial death or inactivity prior to the fermentation process. A systematic evaluation of the experimental procedure identified four major critical factors contributing to fermentation failure: (1) unverified inoculum viability, (2) incomplete anaerobic conditions, (3) potential molasses substrate degradation due to heat sterilization, and (4) the susceptibility of the inoculation protocol to oxygen exposure. These findings provide procedural evaluation guidelines for researchers in designing more reliable ABE fermentation protocols, particularly when using molasses as substrate in laboratories with limited anaerobic facilities.
