Abstract

Avicennia marina and Bruguiera gymnhorriza are plants that have potential as anticancer properties. Previous studies have suggested that Avicennia marina leaf extract has a cytotoxic effect on the stomach whereas Bruguiera gymnhorriza leaf extract has a moderate cytotoxic effect on HepG2 cells. The purpose of this study was to determine the class of compounds and test the cytotoxicity activity of mangrove stem bark Avicennia marina and Bruguiera gymnhorriza against cancer cells MCF7 and T47D. The cytotoxic activity of the extract and fraction was tested against MCF7 and T47D cells using the MTT assay method. The compound yield was obtained from the extraction, fractionation and purification processes. Identification of compounds using NMR. The results showed that the ethanol extract of the Api-api putih mangrove plant contained flavonoids, steroids, terpenoids, phenols, saponins and tannins. While the ethanol extract of tancang mangrove contains flavonoids, terpenoids, phenols, saponins and tannins. The results of purification of Avicennia marina plants obtained compounds which are likely to be triterpenoid derivatives, namely lupeol. Cytotoxic test showed that mangrove stem bark Avicennia marina against T47D cells was active in ethanol extract, semi-polar and polar fractions with their respective values (21.74; 5.26 and 8.78 μg / mL) in the non-polar fraction were moderate with IC50 value 57.88 μg / ml. MCF7 cells gave a moderate category to the semi-polar and polar fractions in Avicennia marina with their respective values (65.60 and 51.84 μg / mL) while the ethanol extract and non-polar fraction were inactive against MCF7 cells with an IC50 value of 105.98 and 169.48 μg / mL. Meanwhile, the purification results of Bruguiera gymnhorriza produced a lupeol compound derived from a terpenoid. Cytotoxic test showed that mangrove bark was susceptible to T47D cells in ethanol extract, polar, semi-polar and non-polar fractions were inactive against T47D cells as well as inactive MCF7 cells.
