Abstract
Mangosteen rind is currently widely used by the public as a health drink. Therefore, it is necessary to standardize the quality of raw materials, extracts and their compounds in order to meet the quality requirements as stated in the Indonesian Herbal Pharmacopoeia. The content of mangosteen rind (Garcinia mangostana L.) consists of various kinds of xantons, especially α-mangostin and γ-mangostin. The existence of these two compounds can be a standard quality parameter of mangosteen rind extract as herbal medicinal ingredients.The standard levels of methanol extract of α-mangostin and γ-mangostin in the mangosteen peel was extracted by HPLC method and the levels of α-mangostin and γ-mangostin from two products on the market were compared. The extract was made by maceration method using methanol. Then fractionated and purified to obtain fractions into isolates by various chromatographic techniques. Identification of isolated compounds by TLCused mobile phase hexane:ethyl acetate (2: 3), H-NMR and HPLC reversed phase with water:acetonitrile (80:20). Quantitative analysis of standard raw compounds using regression equations and compared with compound levels of two related products in the market. The results of the validation analysis of the determination of content for the standard alpha mangostin compound has an average value 0,155 mg/mL whith for A product levels are 0,17 mg/mL and B product levels are 0,23 mg/mL. Content determination validation for gamma mangostin standard compound have an average value 0,386 mg/mL whith for A products are 0,16 mg/mL and B products are 0,232 mg/mL. Validation of the determination method by determining the values of linearity parameters, detection limits and quantitation limits. α-mangostin linearity shows the value of the relation coefficient (r) of 0.993 with a LOD of 0.043 mg / ml and LOQ of 0.129 mg / ml. While γ-mangostin the value of the correlation coefficient (r) of 0.997 with a LOD of 0.169 mg / ml and LOQ of 0.51 mg / ml. α-mangostin and γ-mangostin test results have met the validation criteria
