Abstract

Melinjo (Gnetum gnemon) contains flavonoids and resveratrol which have been shown to inhibit uric acid. This study aimed to determine the inhibitory activity of xanthine oxidase in vitro. The source of the enzyme used comes from the extraction of fresh cow's milk. Inhibition of xanthine oxidase activity by methanol extracts of skin and melinjo seeds in vitro was determined through decreased uric acid production which was monitored with a UV-VIS spectrophotometer at (λ) 290 nm with xanthine as a substrate. The testing of each extract was carried out by making 7 series of concentrations 3.9, 7.8, 15.6, 31.3, 62.5, 125, and 250 ppm. The obtained rate value is then used to calculate the activity value. Then determined IC50. As a comparison allopurinol is used. The results showed that the methanol extract of the skin and seeds of melinjo were able to inhibit the activity of xanthine oxidase in vitro at levels of 100 ppm by 63.53% and 58.90%, while the comparison of allopurinol at 62.5 ppm levels was 80.03%.
