Abstract
Obstacles that often arise in in vitro culture is the presence of contamination by microorganisms that can be prevented by giving PPM (Plant Preservative Mixture) as biocide. Kepok banana’s midrib because it has saponin with the highest content of flavonoids and tannins which have antibacterial and antimicrobial effects so that it has the potential as a biocide material. The purpose of this study was to determine the activity of kepok banana’s midrib powder on in vitro growth of green bean seeds. The parameters are uncontaminated media, plant height, number of roots and number of leaves. The experiment was carried out using a completely randomized design (CRD) with 6 treatments namely positive control, negative control, K1 (0.30%), K2 (0.35%), K3 (0.40%) and K4 (0.45%) , each treatment with 6 replications. Extraction of kepok banana fronds used maceration method which used ethanol solvent 70% (1:10) followed by evaporation to obtain thick extract using watterbath. To get powder, lactose is added with a ratio of 1: 3. The results showed that K4 with 0.45% biocide powder concentration was able to prevent 100% contamination. K4 is more effective as biocide than other concentrations without inhibiting the growth rate of green bean seeds in vitro. Sprout growth was seen from stem height 16.01 cm, number of leaves 2 strands and number of roots 6 strands.
