Abstract
Contamination is an important problem in plant tissue culture techniques. To overcome the contamination of culture media can be done by adding biocides such as Plant Preservative Mixtured (PPM). Kepok banana midribs become bioactive compounds as biocides such as saponins, flavonoids, tannins that can inhibit bacterial growth, accelerate cell growth, and as antibiotics. This study aims to determine the biocide activity of kepok banana midrib powder on the growth of black rice seeds in vitro. The research method used is an experiment with Completely Randomized Design (CRD). Making biocides of kepok banana midrib powder with maceration using ethanol 70% (1:10) for 3 days, and evaporated with a rotary evaporator to get a thick extract at 40oC, extract added lactose powder with a ratio of 1: 3 slowly and stirred until homogeneous . The parameters observed in in vitro culture for 7 days from the process of planting black rice seeds were the percentage of media that did not experience contamination, stem height, number of roots, number of leaves, and seed condition. Kepok banana stem powder with a concentration of 0.45% is more effective than other concentrations and can prevent 100% contamination in the growth of black rice seeds in vitro. The conclusion of this study is the biocide of kepok banana stem powder has the potential as a substitute for PPM in the manufacture of plant tissue culture media in vitro on the growth of black rice seeds with an ideal concentration of 0.45%.
