Abstract

One of the natural product which has anti-cancer activity is avocado (Persea americana Mill.). This study aims to determine the cytotoxic activity of ethanol extract, hexane fraction, ethyl acetate, and ethanol from avocado seeds on WiDr cells. Avocado seed extract was obtained by maceration method with 96% ethanol and then fractionation with n-hexane and ethyl acetate to obtain hexane, ethyl acetate, and ethanol fractions. Cytotoxic tests were carried out using the MTT assay method and the absorbance results were read by ELISA reader. The absorbance data obtained is used to calculate IC50 values. The compound group of the most active fraction was identified using the Thin Layer Chromatography (TLC) method. The cytotoxic test results showed ethanol extract, hexane fraction, ethyl acetate fraction, and ethanol fraction of avocado seeds had an IC50 value of 88.83; 44.04; 159,54; and 510.37 µg / mL against WiDr cells. The results of TLC show a class of compounds found in the n-hexane fraction, including terpenoids and flavonoids. The results of this study indicate that avocado seed extract and hexane fraction has moderate active cytotoxic activity against WiDr colon cancer cells, whereas ethyl acetate and ethanol fractions are not active in cytotoxic against WiDr colon cancer cells.
