Abstract
Theophylline has 3-6 hours half-life of elimination and has a narrow therapeutic window of 5-20 μg / mL . The formation of theophylline formula is expected to produce a stable theophylline levels in the blood that is stable and uniform. The purpose of this research is to know the effect of hydroxypropyl methylcellulose (HPMC) and xanthan gum (XG) matrix combination on physical properties, release pattern, and to get optimum formula that follow zero-order release pattern. Theophylline formulation was performed slowly using a matrix system of HPMC and XG. Formulas of slow release tablets of theophylline were obtained by using a simplex lattice design (SLD) program with two components: HPMC (A) and xanthan gum (B) to obtain F1 formulas (100 mg and 50 mg), F2 (87.5 mg and 62.5 mg), F3 (50 mg and 100 mg), F4 (75 mg and 75 mg), F5 (62.5 mg and 87.5 mg). The measured response is flow velocity, angle of repose, uniformity of weight and content, hardness, friabillity, and dissolution. Based on the physical properties of the five formulas, it is known that the tablet has met the physical properties of the tablet. Xanthan gum is the most dominant factor in increasing tablet flowability and tablet hardness. The drug release was performed in vitro by using dissolution test, based on drug release evaluation it was found that the greater composition of HPMC, the less drug release. Based on expert design calculation , the optimum formula is HPMC: XG (67,53 mg: 82,47 mg)
