Abstract
Doxorubicin is the most active anthracycline chemotherapy agent in the treatment of metastatic breast cancer. The use of doxorubicin is known to be resistant to MCF-7 cells with changes biochemical pathways, those are in metabolic pathways, drug effluents, apoptosis, cell cycle, and DNA repair. Doxorubicin also causes cardiotoxicity due to iron accumulation in the mitochondria. Soursop seeds (Annona muricata L.) are known to be potent as a combination agent of chemotherapy (co-chemotherapy). Development of co-chemotherapy agents with herbal ingredients is expected to decrease the side effects of doxorubicin. The objective of this study was to investigate the cytotoxicity of soursop seeds ethanol extract, their combined effect with doxorubicin against MCF-7 cells and the compounds contained in the extract. Soursop seed extraction using maceration method with 96% ethanol. Cytotoxic assay using MTT assay method and index combination. The parameter obtained from cytotoxic test is IC50, which is concentration which can inhibit cancer cell growth up to 50%. Phytochemical test of soursop seeds ethanol extract using KLT-Densitometry method. IC50 of soursop seeds ethanol extract and doxorubicin to MCF-7 cells were 142,74 μg/mL and 7,41 nM. The combination of soursop seeds ethanol extract with doxorubicin produced a strong synergistic effect on MCF-7 cell with IK value of 0,102 with the optimal concentration of 17,5 μg/mL - 4 μM (⅛ IC50 - ½ IC50) extracts of doxorubicin. Soursop ethanol extract positively contains the alkaloids group
