Abstract
Breast cancer was one of the leading cancers of death. Cancer treatment with a combination of chemotherapy and non-toxic chemoprevention could increase the sensitivity of cancer cells and reduce the toxicity of normal cells. Ashitaba plants (Angelica keiskei) had cytotoxic activity with potent IC50 values in HL-60 cells. 5-Fluorouracil was one of the chemotherapy agents for breast cancer. This was the basis for knowing IC50 of Ashitaba plant and CI (Combinaton Index) from combination of Ashitaba plant with 5-fluorouracil. Identification of chemical compounds used thin layer chromatography with n-hexane: ethyl acetate (7: 3) as mobile phase and silica gel GF254 as stationary phase. The cytotoxic assay used MTT assay with series concentration of ethanol extract of Ashitaba were 500, 250, 125, 62,5 μg/mL and concentration series of 5-fluorouracil were 200, 100, 50, 25 μg/mL. The combination concentration was made from 1/2 IC50, 3/8 IC50, 1/4 IC50 and 1/8 IC50 from extract and drug. The result of KLT show that ethanol extract of Ashitaba leaves contain compounds class of flavonoid, coumarin, phenolic, and terpenoid. Ethanol extract of Ashitaba leaves have moderate cytotoxic activity against T47D cells with IC50 353,86 μg/mL. Cytotoxic test also show that 5-fluorouracil has IC50 against T47D cells was 60,46 μg/mL. The result of combination test of Ashitaba and 5-fluorouracil show that ethanol extract of Ashitaba and 5-fluorouracil have synergistic effect against T47D cells with CI 0,54, 0,42, and 0,28.
