Abstract
Breast cancer is one type of cancer that causes major deaths in the female population in Indonesia. Most cancer therapies cause unwanted effects for patients so many patients are turning to treatment by using materials derived from nature. Methanol extract of the kemukus fruit (Piper cubeba) is known to have cytotoxic activity against breast cancer cells MCF-7, MDA-MB-231, MDA-MB-468. Several previous studies no research has been done on the kemukus fruit of T47D cancer cells, so the purpose of this research is to know cytotoxic activity in methanol extract, butanol fraction, ethyl acetate fraction, and chloroform fraction of kemukus fruit against T47D cancer cells. The maceration method with methanol was selected for extracting kemukus fruit (Piper cubeba). The cytotoxic activity test was performed with an MTT assay. Extracts and fractions that have less IC50 values were tested the content of compounds with TLC (Thin Layer Chromatography).. The result of cytotoxic activity test showed that IC50 methanol extract and chloroform fraction of kemukus fruit (Piper cubeba) were 134,95 μg / mL and 225.3 μg / mL, respectively butanol fraction and ethyl acetate fraction show that did not have cytotoxic activity. The result of TLC showed that methanol extract and chloroform fraction contain secondary metabolite compound of alkaloids, lignans, and terpenoids.
