Abstract
Culture is one of the techniques of plant propagation in aseptic conditions. The main problem in plant propagation techniques of plant net culture is contamination by bacteria or fungi, to overcome this contamination often uses Plant Preservative Mixture (PPM) as a biocide. The ambon banana stem and rind contains chemical compounds such as saponins, tannins, flavanoids, anthraquinones and quinones that serve as biocides. The purpose of this study was to determine the ratio of potential biocide between the stem extract and the banana rind to prevent the contamination of in vitro culture. This research method used Randomized Complete Design (RCD/RAL) with two factors, factor 1, that is, the origin of ambon banana stem extract (E1), the banana rind of Ambon (E2) and factor 2 is concentration: 0.5% (K1) And 1% (K2). The extraction of banana stemand rindused maceration method. The results showed that in the bottle of banana rind extract was more inhibited contaminant by 80% and normal growth of sprouts, whereas in the percentage of uncontaminated stem extract was high and abnormal growth of bean sprouts. The banana rind extract at 1% concentrations is more effective to prevent the contamination of green beans seed in vitro.
