Abstract

Green betel leaves (Piper betle L.) are used and believed to be an alternative medicine in treating various types of diseases. This study aims to determine the antibacterial activity of ethanol extract of green betel leaves against Staphylococcus epidermidis and Escherichia coli bacteria and to identify groups of compounds that have antibacterial activity in ethanol extract of green betel leaves. Extraction of green betel leaves (Piper betle L.) using 96% ethanol solvent with maceration method. Antibacterial activity test was carried out using disk diffusion method, identification of groups of compounds that have antibacterial activity was carried out using bioautography test. The levels of ethanol extract used were 2 mg/disk, 4 mg/disk, 6 mg/disk, and 8 mg/disk. Positive controls using Erythromycin and Ciprofloxacin and 50% DMSO solvent was used as negative control. The results of this study indicate the antibacterial activity of ethanol extract of green betel leaves against Staphylococcus epidermidis with a concentration series of 2 mg/disk, 4 mg/disk, 6 mg/disk, and 8 mg/disk producing an average inhibition zone of 9.5 mm; 12 mm; 15.12 mm; and 15.62 mm. While the results against Escherichia coli with a concentration series of 2 mg/disk, 4 mg/disk, 6 mg/disk, and 8 mg/disk produced an average inhibition zone of 10.25 mm; 15.12 mm; 15.87 mm; and 16.25 mm. The results of the TLC test showed that green betel leaves contain phenol, saponin and alkaloid compounds. The results of the bioautography test showed that the compounds that have antibacterial activity against Staphylococcus epidermidis are phenol compounds at Rf 0.8 and the compounds that have antibacterial activity against Escherichia coli are alkaloid compounds at Rf 0.7.
