Abstract

Sea cucumbers are marine organisms found in many waters of Indonesia which are included in the phylum Echinodermata. One species used as an anticancer is the sea cucumber Holothuria atra Jeager, 1833. This study aims to isolate and identify secondary metabolites and to determine the anticancer activity of the Holothuria atra in vitro on HeLa, T47D, WiDr and Vero cell lines. The active compounds was elucidated with spectroscopic analysis by using UV, FT-IR, LC-MS, and 1H-NMR. Anticancer testing activities were performed by the MTT assay. Flowcytometric analysis was performed to investigate cell cycle and induction of apoptosis. Double staining was performed to observe the morphology of cells went apoptosis performed on cells. The results showed that the sea cucumber of Holothuria atra indicated contain active compound of Holothurin B1. Based on the LC-MS spectra, the compound has the molecular formula C41H65NaO16S (m/z 868.75). Holothurin B1 has cytotoxic activity against HeLa, T47D, WiDr and Vero cells with IC50 values of 137.37; 87.67; 99.57 and 1878.76 μg/ml. Analysis of T47D cell-cycle in active compounds with concentrations of 87 and 43.5 μg/ml was able to induce cell-cycle arrest in the sub-G0/G1 phase. The results of flowcytometry showed that most of the cells (97% at concentration of 87 μg/ml and 84.58% at concentration of 43.5 μg/ml) had induce apoptosis and proven by observing cell morphology with double staining. Based on cytotoxic test, cell cycle analysis and apoptosis induction test, we can infer that the Holothurin B1 is potential to be developed as anticancer agent.
