Lately, there are many studies that examine the transmission and spread of HIV virus which focus on human behavior, the risks and dangers of the disease also how the transmission happened. However, the availability of information and the study of how the disease spread which associated with spatial dimension has not been widely available. One of analysis that can illustrate the spatial spread of HIV sufferers is the Hot Spot Analysis (Getis-Ord Gi*) in ArcGis software which use all number of HIV sufferers data for analysis. The method which had been using in this research is quantitative-analytic that use mathematical formula or statistics by utilize Hot Spot Analysis (Getis Ord Gi*) feature in ArcGIS software to obtain the results of the hot spot spread or the main clustering of HIV sufferers in Sorong City 2016. In addition, qualitative-descriptive method also used to analyze the results of hot spot analysis reinforced by indepth interview to obtain correlation of prostitution location and localization to hot spot spreading of HIV sufferers. The results of this study indicate that; (1) The hot spot spreading of HIV sufferers in Sorong City on 2016 is located in the central region of Sorong City because most HIV sufferers are in that region, with the subdistrict region which become hot spot area are Malanu, Klagete, Malaingkedi, Sawagumu and Klawuyuk with the confidence number to be hot spot area reaching 99%, Remu Utara and Klasabi with the confidence number to be hot spot area reaching 95% and Klawalu with the confidence number to be hot spot area reaching 90%; (2) The existence of prostitution localization that named Lokasi Malanu is very related with hot spot spread of HIV sufferers in Sorong City on 2016.
