Abstract

Diclofenac sodium is a non steroidal antiinflammatory drug that has therapeutic effects as an antipyretic. The production of sustained release tablet is needed the matrix to prolong the time to keep of the drug in the body and control the extrication of the drug. The purpose of this research is to know the effect of the eudragit combination (ED) and xanthan gum (XG) on the physical characteristic of the granule and the tablet as well as the drug release profile. The sustained release tablet of diclofenac sodium of this research is made in four formulas with the concentration ratio of eudragit (ED) and xanthan gum (XG) were the first formula was (3%w/w : 1%w/w); the second formula was (1%w/w : 1%w/w); the third formula was (1%w/w : 2%w/w); and the fourth formula was (3%w/w : 2%w/w). The evaluation of the physical characteristic of granules and tablets include the flow speed, angle of repose, tapped density, variety of weight, hardness, friability, the uniformity of content, and dissolution. The data was from the evaluation of the physical characteristic of granules and tablets were processed by Design Expert 11.0.5.0 of the factorial design model then analyzed by One sample T-test with 95% of confindence level. The results show that more ED can decrease hardness of the tablet and dissolution speed while more XG can increase the flow speed, angle of repose and tapped density, hardness of tablet, and the dissolution speed. The optimum formula was gotten by the concentration comparison ED:XG (2,68%w/w : 2%w/w) with desirability of 0.691.
