Abstract
Immobilization of enzyme is attachment enzyme both physically and chemically to an insoluble support solid in water. Immobilization of enzymes can be done in various ways, one of them is absorption on matrix with chitosan or also called crosslinking. The purpose of this research is to know optimum conditions in immobilized enzymes. The enzyme used is papain enzyme. Crosslinking agent used is CaCl2. Variables for immobilized papain enzyme are pH, time and concentration of enzyme. The pH range for immobilization of papain enzyme is 6-9, the time is 3-12 hours and the concentration is 10-40 mg/mL. The optimum conditions for immobilized papain enzyme were at pH 8, immobilization time 6 hours and concentration of enzyme 40 mg/mL.
