Abstract

Problems which often appear in in vitro cultures are contamination caused by bacteria or fungi. Generally, contamination is able to be prevented by using PPM (Plant Preservative Mixture) as an antimicrobial. One of the benefits of leaves and fruits belimbing wuluh can be used as antimicrobial contain of saponin, flavonoids, tannins, and triterpenoids. The purpose of this research is to determine the potential comparison of biocide between fruit extract and leaves extract of belimbing wuluh to prevent contamination of in vitro cultures. The research design uses a complete randomized design (RAL) of 2 factors: factor 1: bilimbi’s leave (B1); bilimbi’s fruit (B2) and factor 2: 0,5% extract concentration (K1); 1% extract concentration (K2). Fruit extract and leave extract of belimbing wuluh by maceration method using ethanol solvents 96%. Parameters used for 7 days are sproud condition, amount of leaves, stem height, and percentage of uncontaminated media. The percentage of uncontaminated media on treatment of B1K1, B1K2, B2K1, dan B2K2 are 80%, 60%, 100%, 100%. Sprout condition in B1K1 dan B2K1 is normal, while B1K2 dan B2K2 is abnormal. Leaves extract 0,5% of belimbing wuluh effective as biocide in in vitro culture than fruit extract of belimbing wuluh.
