Abstract
The main topic in the in vitro culture is contamination of microbes such as bacteria or fungus. In order to avoid the contamination, the PPM (Plant Preservative Mixture) is mostly used as biocide. Roots and stem of ambon banana contains flavonoid, tannin and saponin type of compound which have a lot of use such as a biocide. The aim of this research is to understand the comparison of biocide potential the roots and stem extract of ambon banana to avoid the contamination in the in vitro culture. The research used experimental method with completely randomized design using two treatment factor is origin extract (roots and stem of ambon banana) and extract concentrations (0,3% and 0,6%). The roots and stem extraction of ambon banana used with maceration method with methanol solvent (1:5). Next, the maceration filtrate result is distillated with 650C temperature. The parameter which is observed in 7 days from the growth of peas is the percentage of uncontaminated medium, stem height, number of roots, number of leaves, and peas condition. The research results show that extract of ambon banana’s stem with 0,3% concentration is the most effective to avoid the contamination as much as 100% without slowing down the growth of peas in the in vitro culture and peas condition in all treatment is normal.
