Abstract

Ginger (Zingiber officinale) is a spice with potential as a medicinal plant that has significant economic significance globally. The economic value of ginger lies in its rhizome, which contains many bioactive compounds that are beneficial for health. Emprit ginger (Zingiber officinale var Amarum) has the highest gingerol content compared to other ginger varieties. Herbal medicines contain a complex mixture of compounds, so standardization is carried out using marker compounds. Gingerol (C17H26O4) is the main compound in ginger that is most active and abundant, with 6-gingerol as the most dominant form. This study aims to vary the extraction methods, namely maceration, reflux, UAE (Ultrasonic Assisted Extraction), and MAE (Microwave Assisted Extraction) with 70% ethanol solvent. Isolation of the 6-gingerol compound in this study used the preparative TLC (Thin Layer Chromatography) method. Confirmation of the 6-gingerol compound using 1H-NMR (Nuclear Magnetic Resonance) spectroscopy analysis and maximum wavelength measurement using a UV-Vis spectrophotometer. The results showed that the most optimal extraction method was the reflux method, as indicated by the spot thickness in the TLC analysis. The review process using preparative TLC proved effective in isolating the 6-gingerol compound. The results of the 1H-NMR analysis showed agreement with the 1H-NMR spectrum data of the 6-gingerol compound reported in the literature and the maximum wavelength measurement showed agreement with the reference data, namely at a wavelength of 281 nm. Thus, the results of the isolation from ginger are suspected to contain the 6-gingerol compound.
