Abstract
Kaffir lime is widely used in research and commercial applications and is reported to have various pharmacological activities that support its use as a raw material for herbal medicines. Herbal products require marker compounds as a reference for quality control. Hesperidin is the main flavanone in kaffir lime peel which is listed as a marker compound in the Indonesian Herbal Pharmacopoeia. This study aims to determine a rapid extraction method to obtain marker compounds and identify the isolated compounds as standard compounds. The extraction methods used include maceration, reflux, Microwave Assisted Extraction (MAE), and Ultrasonic Assisted Extraction (UAE) with methanol as the solvent. Isolation was carried out using column chromatography and preparative Thin Layer Chromatography (TLC). Confirmation of the compound structure used 1H-NMR (Nuclear Magnetic Resonance) spectroscopy analysis and measurement of the maximum wavelength (λmax) using a UV-Vis spectrophotometer. The 1H-NMR analysis results showed agreement with the 1H-NMR spectrum data of the hesperidin compound and the maximum wavelength measurement showed agreement with the reference data, namely at a wavelength of 285.5 nm. Based on this, the results of the isolation of kaffir lime peel are suspected to contain hesperidin compounds.
