Abstract

Infection is a disease that can cause clinical symptoms or asymptomatic triggered by microorganisms such as bacteria. Pseudomonas aeruginosa and Bacillus subtilis are bacteria that can cause nosocomial infections. The purpose of this study was to determine the antibacterial activity of extracts and fractions of white galangal rhizome (Alpinia galanga) against the Pseudomonas aeruginosa and Bacillus subtilis and identify the class of compounds that act as antibacterial agents. Extract obtained from maceration of galangal using 70% methanol. Disc diffusion method was chosen in the antibacterial activity test. The samples tested were extracts with a concentration of 10.5; 12; 13.5; and 15 mg/disc, methanol fraction 15 mg/disc, ethyl acetate fraction 15 mg/disc, chloroform fraction 15 mg/disc, chloramphenicol as positive control, and DMSO as negative control. The results of the methanol extract test showed antibacterial activity against Pseudomonas aeruginosa and Bacillus subtilis at a concentration of 15 mg/disk with inhibition zone diameters of 13.5 ± 0.41 mm and 16.0 ± 0.41 mm. The results of the ethyl acetate fraction test showed the greatest antibacterial activity against Pseudomonas aeruginosa and Bacillus subtilis with inhibition zone diameters of 12,0 ± 0,41 mm and 13.7 ± 0,24 mm. Identification of white galangal rhizomes (Alpinia galanga) compounds using the TLC method with silica gel GF254 as the stationary phase, chloroform : ethyl acetate (1:1 v/v) as the mobile phase to elute the extract plate, and chloroform : methanol (9:3 v /v) as the mobile phase to elute the ethyl acetate fraction plate. Phytochemical test results of white galangal rhizome (Alpinia galanga) showed the methanol extract positively contained alkaloids, phenolics, flavonoids and terpenoids, while the ethyl fraction positively contained phenolics, flavonoids and terpenoids. Bioautographic tests showed that the class of compounds capable of inhibiting Pseudomonas aeruginosa and Bacillus subtilis bacteria were phenolics, flavonoids, and terpenoids.
