Abstract

Lipase enzymes are enzymes that hydrolyze lipids, triglyceride esters, to form fatty acids and glycerol. Utilization of Aspergillus niger is a potential source of extracellular lipase because it is able to produce lipase enzymes with high activity and levels, easy maintenance, economical cost, easy to obtain, and can grow on relatively inexpensive media. This study aims to determine the optimum nutrition in the solid fermentation process using rice bran substrate and Aspergillus niger strain. The method used in this study is solid fermentation and analysis using the titrimetric method with the NaOH and Lowry methods to determine the protein content. The results showed that the best activity for lipase was ammonium sulfate with the smallest variation of 30% and 2 ml of a mixture of 95 ml amyl alcohol and 5 ml isopropanol resulted in an activity of 0.62 U/mL. Then the best concentration of protein content was ammonium sulfate with the smallest variation of 30% with 1 ml of a mixture of 95 ml of amyl alcohol and 5 ml of isopropanol to produce a concentration of 0.18 mol
