Abstract

The grade C soybean has always getting judgment as a compost heap and is barely used. Nevertheless, it also has a high nutritional value, such as protein. Therefore, it can be utilized as a substrate in the production of protease enzyme through the process of solid fermentation the grade c soybean with inoculum tempeh Kediri. The Protease enzyme has a high economic value because of its efficiency and it's also environmentally friendly, which can contribute to protecting the environment by taking advantage of solid waste. This research has a purpose to understanding the effect of moisture content with variances 85%, 80%, 75%, and the nutrition additional like Magnesium chloride with variances of 10ml, 20ml, 30ml toward the activity of protease enzyme that produces the Rhizopus enzyme. The experiment activity of the protease enzyme was done on a spectrophotometer with casein as the substrate. The result of protease enzyme activity shows the diversity of value activity. In the range of variables comprehended that the condition of the optimum environment with the activity of protease enzyme is 75% of moisture content and additional nutrition in amounts of 30mg, the activation of protease enzyme obtained of 2,0949 unit/mL and the lowest protease enzyme activity obtained of 0,9061 unit/mL, while the control sample obtained protease enzyme activity of 1,2186 unit/mL.
