Abstract

Cervical cancer is a disease with a high mortality rate in the world. One of the natural ingredients targeted is Spirulina platensis. This study aimed to determine the cytotoxic effects of ethanol and methanol extract of Spirulina platensis on HeLa cells. Extraction of Spirulina platensis was carried out using maceration method with ethanol 96% and methanol. Cytotoxic activity tests were carried out using the MTT assay method with a 500 concentration series; 250; 125; 62.5, and 31.5 µg / mL. Cytotoxic test results in the form of absorbance data are read on the ELISA reader which is then used to calculate the average percent of living cells. From this study, the average percentage of living cells at a concentration of 500 µg / mL of Spirulina platensis ethanol and methanol extract was 72.667% and 81.901%. The average yield of living cells is still very high above 50%, so IC50 cannot be calculated. The results of this study concluded that ethanol and methanol extract of Spirulina platensis did not have cytotoxic activity. Identification of compound content using Thin Layer Chromatography (TLC) with the mobile phase of ethyl acetate: n-hexane (7: 3) and silica GF 254 phase. Ethanol extract Spirulina platensis is thought to contain flavonoid and steroid compounds. While the Spirulina platensis methanol extract contains chlorophyll α as well as flavonoid and steroid compounds.
