Abstract

Bawang Dayak (Eleutherine americana Merr.) have been used by the community in the form of powder, simplicia or decoction, in order to make it easier to use thus it is formed in tablet. The improper combination of binding agent (gelatin) and disintegration agent (explotab) can cause the tablet not to be optimum, so formula optimization needs to be done. This study aims to determine the optimal combination of gelatin and explotab based on the physical properties and their antioxidant activity. Extract of bawang Dayak was obtained from maceration by using 96% ethanol. Eight tablet formulas were made with lower and high level of gelatin concentrations (7:21 mg), low and high level of explotab (14:28 mg) using Factorial Design method analyzed by Design Expert 11 (trial) to obtain optimum formula. The optimum formula was verified and analyzed by using SPSS Software one sample t-test with 95% confidence level. The antioxidant test of bawang Dayak extract was done by using DPPH method (2,2-diphenyl-1-picrylhydrazyl) with UV-Vis spectrophotometer at λ 516 nm. The results show that combinations of gelatin and explotab levels can improve the flow rate, angle of repose, tapped density, hardness and the tablet friability, but it decreased weight uniformity and the disintegration time of tablets. The results of the optimum formula were obtained by a combination of gelatin:explotab each of (11.62:23.45 mg). Bawang Dayak extract IC50 the value was 59.414 µg/mL, while % inhibition of bawang Dayak extract tablets and placebo tablets was 45.771% and 1.220%. Antioxidant activity of bawang Dayak is not affected by the combination of tablet excipients.
