Abstract

Theophylline is an effective asthma remedy for chronic bronchial asthma therapy and bronchospastic reactions. Its narrow therapeutic index is 10- 20 mcg / ml so the risk of toxicity is high. The matrix system is used to control drug release in the sustained release tablet to maintain the therapeutic levels of the drug over a long period of time. This research aims to determines the influences of NaCMC and ethylcellulose combination on granule flow properties, tablet physical properties, and dissolution profiles. The sustained release tablets of theophylline were made in five formulas by wet granulation method (FI (30 : 15), F2 (15 : 30), FIII (22,5 : 22,5), FIV (26,25 : 18,75), FV (18,75 : 26,25)). Evaluations were perform the flow rate, angle of respose, hardness, friability, weight uniformity, drug content and dissolution. Data were analyzed by Design Expert 11 (trial) and SPSS 23 program with 95% confidence level. The results of this study show that combination NaCMC and ethylcellulose decrease granule flow properties, decrease weight uniformity, increase friability, decrease drug content, and increase dissolution rate. The optimum formula was obtained from the concentratio ratio of NaCMC : Ethylcellulose (28,87 mg: 16,12 mg).
