Abstract

Decolorization of Brillilant Blue R (RBBR) Remazol can be carried out enzymatically. The RBBR dye-polarization enzyme can be obtained from the Pleurotusostreatus and Lentinulaedodes (shitake) fungi. Enzymes produced by pleurotusostreatus include lacase, manganese peroxidase (MnP) and lignin peroxidase (LiP) enzymes. RBBR decolorization activity test was performed by extracting ligninolytic and lignosellulatic enzymes from Pleurotusostreatus and shitake fungi. Mushroom mycelium was developed on a PDA medium, after 6 days of mycelium inoculated on palm stem waste substrate by adding nutrients to trigger early growth of the fungus. To determine the effect of nutrition and pH on activity of enzyme decolorization experiment was done by giving CuSO4 nutrition equal to 0,025; 0.05; 0.075 g/L and conditioned in an environment with a pH of 4; 5; 6. After 6 days the enzyme is extracted by adding the citrate buffer then centrifugation at 10 minutes at 2000 rpm. The enzyme extract was then applied to the RBBR dye and incubated for 1 hour at 45oC. The activity of decolorization by enzyme is determined by the decrease of absorbance value of RBBR. The highest enzyme activity of pleurotusostreatus fungus was achieved at 0.075 g/L CuSO4 concentration and at pH 4. The addition of CuSO4 was able to trigger the secretion of lacase enzyme from fungi. While in shitake, the highest activity was achieved at 0.025 g/L CuSO4 concentration at pH 5.
