Abstract

Newcastle Disease is kind of a viral envelope who has two membrane proteins, the hemagglutinin-neuraminidase (HN) protein which is associated with cell attachment and release, and the fusion protein (F) which mediates the viral envelope with the cellular membrane (Uddin et al., 2017). Newcastle Disease infection in poultry is characterized by pathological abnormalities in the humoral (bursa fabricius) and cellular (thymus and spleen) lymphoid organs. These pathological abnormalities can affect the work of the specific and non-specific immune systems in inhibits Newcastle Disease virus infection. Compound by ethyl 4-(4-hydroxy-3-methoxyphenyl)-6-methyl-2-oxo-1,2,3,4 tetrahydropyrimidine-5-carboxylate can be used as an antiviral. This study has purpose to determine the effect of activity of the compound by ethyl 4-(4-hydroxy-3-methoxyphenyl)-6-methyl-2-oxo-1,2,3,4 tetrahydropyrimidine-5-carboxylate when it use to Newcastle Disease virus, which was inoculated in embryonic chickens. The method used in this study was an experimental method using 13 embryonic chicken eggs which were divided for 4 groups, that was 1 control groups and 3 treatment groups. The test used is the hemagglutination test, this method is to determine the virus titer. Virus titer can be determined by looking at the last well that shows positive hemagglutination, that is, the well with the largest dilution factor of the isolates where hemagglutination occurs. The microtiter on the microplate calculated by the inhibitory formula can show the percentage of infection inhibition. The value obtained at a concentration of 1 µg/mL was 0% in I, II, III replications. Then, at a concentration of 10 µg/mL obtained 86.621% in the third replication. While in the concentration group of 100 µg/mL obtained 86.621% in replication I; . 99.951% on replication II; 99.609%. in replication III. The results showed that the administration of the compound ethyl 4-(4-hydroxy-3-methoxyphenyl)-6-methyl-2-oxo-1,2,3,4-tetrahydropyrimide-5-carboxylate was effective in inhibiting the Newcastle Disease virus at a dose concentration of 100 µg/mL.
