Abstract

Skin infections can be caused by pathogenic bacteria Pseudomonas aeruginosa and Staphylococcus aureus. The high incidence of infectious diseases accompanied by an increase in bacterial resistance to antibiotics encourages the use of herbs that have antibacterial activity, one of which is widuri leaf (Calotropis gigantea). The purpose of this study was to determine the antibacterial activity and to identify the active compounds from Calotropis gigantea against Pseudomonas aeruginosa and Staphylococcus aureus. The antibacterial activity of Calotropis gigantea leaf extract was tested using the disk diffusion method with a concentration of 7,5 mg/disc; 8 mg/disc; 8,5 mg/disc dan 9 mg/disc. The results showed that the leaf extract of Calotropis gigantea possesed antibacterial activity against Staphylococcus aureus at the concentrations of 7,5 mg/disc; 8 mg/disc; 8,5 mg/disc dan 9 mg/disc resulting in the zone of inhibition of 9,25 ± 0,35; 9,75 ± 0,35; 10,0 ± 0,0 and 10,5 ± 0,0 respectively. Meanwhile, at the same concentration there was no inhibition against the growth of Pseudomonas aeruginosa. The content of active compounds in the leaf extract of Calotropis gigantea was identified using the TLC method with the mobile phase of chloroform: benzene: methanol (5: 3: 2) v/v/v. TLC results showed that the extract of Calotropis gigantea contained flavonoids, alkaloids, tannins and terpenoids. Bioautography test results showed that terpenoid compounds from methanol extract of widuri leaves (Calotropis gigantea) had antibacterial activity against Staphylococcus aureus.
